Just a heads up, it could be a bad port, but a lot of the time when there is a "bad port", it is not the port itself but actually damage to other locations on the motherboard which are responsible for accessing that port. So you might fix it by replacing the port (if that is even possible), or you might replace the port and find it still doesn't work.

+ Very good, accurate answer. In fact if the slot/port itself were bad that would mean it has bent pins or has corrosion/residue on them preventing good, proper contact with the RAM stick. Unless the pins are bent, a good cleaning will resolve the problem. Aside from that. unusually the problem is actually in the slot's power circuit and not the data bus. Many times it is caused by people not paying attention to the specified voltage for the slot and putting RAM in it that requires more voltage than the slot was designed to handle or liquid damage. Some slots are designed for 1.5v others for 1.65v, 1.8v, 2.0v, etc.... Just because RAM will work in the slot doesn't mean it is the right RAM or it can't hurt the slot's circuitry over time. Just because RAM is Apple approved doesn't mean it is approved for your Apple computer.

i had this problem on my Powerbook 15 1.65 ghz but it was an issue with apple. Unfortunately my machine did not fall into their serial number recall list but had the exact problem, work that out. Anyway, the repair was supposed to be a motherboard replacement and apple quoted 1200€ here in germany, more than what the 1.5 year old powerbook was worth at the time but when i searched online I found a company called werner media in the US and they fixed the machine for 200$. It worked a treat for a long time but then when I updated the Operating saystem from panther to tiger the ram slot stoped working again. I have since given the now 5 year old powerbook to my son adn installed leopard, it runs well and sometimes when he starts it he had 2gb ram and other days he has 1gb ram so basically the trick is to find out what they at werner media do and then once fixed...do not change the OS.

I always buy the extra warranty for my apple laptops now (3 years) and I don't regret it as I have had my display on my second mac replaced twice which was also a very expensive repair. It just sucks that you have to pay 250$ for a warranty which is really something that should be standard.
