I believe that the problem is with the main mic near the charging port. Can anyone help with replacing or repairing the mic and/or the wiring. Total novice so all help gratefully received.

Alisha is correct. Your iPhone has 3 microphones, one at the bottom for phone calls NOT ON speakerphone, one inside of the front earpiece speaker mesh for calls when USING speakerphone, and one on the rear of the phone near the camera for audio for videos, etc.
If you people can’t hear you but you can hear them, it is one of these 3 microphones most likely.
Do a process of elimination essentially by calling someone on speakerphone and see if that fixes the problem. Then taking a video. Then taking a voice memo. This would test all the mics.
The video will use the back microphone. The voice memo will use the bottom mic same as the NON Speakerphone calls, and finally the SPEAKERPHONE call will test the 3 and final mic in the earpiece speaker.
use these diagnostic elimination steps to determine with which microphone the problem lays.
If it is the mic at the bottom at fault, a new lightning port assembly is in order, as the bottom mic is built into the lightning port assembly.
FYI! If you make any repairs on this device you make, don’t forget to purchase the replacement display adhesive so that you can get your water and dust protection back after opening the device! It only costs a few bucks and is worth every penny!

It could be the Mic, but it could be the board. These are notorious for audio issues. Can you put a call on speakerphone? Record a voice memo? I would see if other functions work because this board issue is so prevalent.
